Mechanism-based analysis of drug combination effects on HCV replication in Huh-7 cells

Abstract


The combination of drugs is a common practice for enhancing the efficiency of drug treatment and
reducing the risk of emergence of drug resistance. However, selection of the optimal combination and the
optimal doses remain poorly understood. Here we tried to find an optimal drug combination among four kinds
of anti‐HCV drugs (IFN‐α, telaprevir, cyclosporine, and halopemide) based on mathematical model which
describes HCV replication in Huh‐7 cells. Interestingly, we found that only combinations which include
halopemide achieve synergistic response to the HCV replication. In this talk, I will explain our in vitro
experiments and their drug efficiency analysis based on mathematical model. Furthermore, I will discuss about
possible mechanism which consistently explains all combination response.
